In a prospective cohort of 142 patients with transthyretin cardiac amyloidosis from Columbia University, unsupervised machine learning applied to plasma proteomics profiling of 7289 proteins identified three molecular subtypes (subtype A: n=49, subtype B: n=38, subtype C: n=55) that showed different survival risks during a median follow‑up of 5.2 years (log‑rank P=0.035) without significant differences in traditional clinical stages. An ensemble machine‑learning model using transcriptomic profiles of circulating CD14+ monocytes achieved a diagnostic AUC of 0.888 in 567 independent samples and a recurrence detection AUC of 0.975 in 51 postoperative samples. In 30 glioma patients, lower postoperative model scores were significantly associated with longer progression‑free survival (log‑rank P = 0.034). [2]
